Deep phenotypic and functional characterization of NK cells during NK-mediated cytotoxicity

Abstract Natural Killer (NK) cell-mediated cytotoxicity is a critical mechanism of the immune system to respond against harmful stimuli. The development emerging cell therapies, such as CAR-NK cells, requires deeper characterization cultured NK cells. Here we show phenotyping ex vivo and in vitro demonstrate how high quality reliable reagents from BioLegend ensure more depth cytotoxic analysis. cells were isolated fresh human PBMCs, using our MojoSort™ Human Cell Isolation Kit, incubated at different ratios with Tag-it Violet™ stained K562 target IMDM + BioLegend’s Cell-Vive™ T-NK Xeno-Free Serum Substitute, presence CD107a-PE. Cells then harvested for surface supernatants collected cytokine analysis LEGENDplex™ CD8/NK Panel. Apotracker™ Green staining significant decrease proliferation increase apoptosis, respectively.
